The premature activation of digestive proenzymes (zymogens) within the pancreatic acinar cell is an early and critical event during acute pancreatitis. Our previous studies demonstrate that this activation requires a distinct pathologic rise in cytosolic calcium. However, the mechanism of this calcium effect remains unclear. An important target of calcium in acinar cells is the calcium-calmodulin dependent phosphatase calcineurin (PP2B). While PP2B appears to have a role in regulated amylase secretion, its effect on zymogen activation is unknown.
