It is still unclear on the expression of cancer-testis antigen genes in glioma stem cells.OBJECTIVE:
To detect the expression of cancer testis antigen genes in glioma stem cells.METHODS:
Stem cells were cultured and isolated by neurosphere culture methods in DMEM/F12. Then the expression of cancer testis antigen genes were analyzed among parent cells, stem cells and differentiated cells by using semiquantitative RT-PCR and real time PCR.RESULTS AND CONCLUSION:
We isolated tumor spheres from glioma cell lines by neurosphere culture. And then tumor spheres were cultured in medium containing serum, the morphology of the tumor spheres changed to adherent and they were not distinguishable from the parent cells. The expression of cancer testis antigen genes were analyzed by semiquantitative RT-PCR and quantitative RT-PCR and it was found that the expression were highest in stem cells compared with parent cells and differentiated cells. Cancer testis antigen genes may present as surface antigens in cancer stem cells and these findings suggest that cancer testis antigen genes may be helpful for targeted vaccine therapy against cancer stem cells in glioma patients.
