Abstract

BACKGROUND:

Previous study shows that expression of peroxidase 6 in fetal rat testis increase significantly induced by in utero exposure to di-n-butyl phthalate.

OBJECTIVE:

To further investigate the effect of prenatal di-n-butyl phthalate exposure on peroxidase 6 expression.

METHODS:

Rats of 14 to 18 days of pregnancy were exposed to di-n-butyl phthalate with the dose of 800mg / (kg•d). Testes were harvested from the fetal rats from gestational day 21 and peroxidase 6 was extracted. The expression of peroxidase 6 was detected and analyzed by western blot and immunohistochemical methods.

RESULTS AND CONCLUSION:

According to western blot, the expression of peroxidase 6 in experiment group (0.205±0.020) was significantly higher than that in the control group (0.110 ± 0.020, P < 0.05). According to immunohistochemistry, peroxidase 6 was mainly localized in Leydig cells of fetal rat testes. These findings indicate that peroxidase 6 may antagonize di-n-butyl phthalate toxicity by antioxidation and stimulating the proliferation of Leydig cells. The function of peroxidase 6 needs further research.
