Abstract

BACKGROUND:

A strontium incorporated freeze dried bone material has been developed in previous report.

OBJECTIVE:

To evaluate the cellular biocompatibility of a novel strontium incorporated freeze dried bone material in comparison with normal material.

METHODS:

MC-3T3 osteoblasts were inoculated on the surface of strontium incorporated freeze dried bone material and normal freeze dried bone material, and the cell proliferation, alkaline phosphatase activity and osteocalcin activity were evaluated and cell micro morphology were observed by scanning electron microscope.

RESULTS AND CONCLUSION:

Osteoblast grew normally on the strontium incorporated freeze dried bone material. On days 3 and 5, cell quantity on the strontium incorporated freeze dried bone material was significantly higher than that on the normal freeze dried bone material (P < 0.05). The alkaline phosphatase activity on the strontium incorporated freeze dried bone material was significantly higher than that on the normal freeze dried bone material (P < 0.05). But there was no significant difference of osteocalcin activity between two groups (P > 0.05). On the surface of strontium incorporated freeze-dried bone material, the osteoblast spread well. Cells were polygonal or spindle-shaped with abundant cytoplasm and the pseudopodia closely attached to the material surface. Some micro filopodia could be observed at the end of cell protrusion. Strontium incorporated freeze dried bone material had excellent cell compatibility, and the growth of osteoblasts on its surface was superior to those on the surface of normal freeze dried bone material.
